Alignment: do my wheels have 2 come off

Alignment: do my wheels have 2 come off
At anytime during an alignment do the wheels have to come off, or any kinda device be placed on the wheels. (excuse my lack of knowledge) thanks in advance

wheels stay on.
At a shop-
Device attaches to wheels. Usually some sort of laser device is clamped onto the rim
At the races, or at home in the garage,
we use 2 coke cans and a tape measure
You can probably guess which one is more accurate.
